If you have enough memory that you can slurp the files into arrays, you might consider slurping them into hashes instead. Then checking if a number is on the list will be very fast, I.E.
if( $do_not_call{ $test_number } ){
print "Do Not Call $test_number\n";
}
As far as database options, you might consider DBD::SQLite. It's very fast.
